I am going to be taking classes from Straighterline, LSU, BYU, ISI (Idaho) and other places. I want to get a GPA on my transcript for graduate school reasons. Which one of the big 3 will calculate GPA based off these courses?

It's highly unusual to have a school allow GPA points for transferred courses. You can check the transfer section on the 3 websites in just a few minutes. You can do that for dozens of schools. If you come up with a short list you could post it here. TESC for sure does not.
Also, know that SL uses a pass/fail, so no GPA points there.

One of the unusual benefits at Excelsior is that they do transfer grades and calculate a GPA for them. It includes outside courses, Ohio U exams, and some (non-refreshed) DSST exams. They also have a second GPA for Excelsior courses and ECE exams. I think UExcel exams are also graded.
I am not aware of their awarding grades for StraighterLine, but COSC does give grades for SL according to posts I have read. COSC also awards grades for some DSST exams. Check burbuja0512's signature for more details about COSC.
