TESC will accept transfer credits that existed before you enroll with them, as well as during your enrollment. A lot of people complete some or all of their CLEP/DSST/etc. credits before enrolling to ensure they are out of the way to finish within the enrollment period, but you do can do it the other way around if you choose.
You technically do not NEED to take a course for a GPA - people have graduated without a GPA - but most of us want to be able to list that on resumes, etc. That course must be taken directly through the school, and to earn a letter grade, must be either an Online or Guided Study course type. In other words, you cannot use e-Pack, TECEP, or any of the other course types as they are all Pass/Fail. It will go towards your residency requirement, though. Online and Guided Study courses are on fixed 12-week schedules, so consider that when mapping out your timeline.
